This variant lies in the CEACAM16 gene (transcript NM_001039213.4) at coding-DNA position 160, where T is replaced by C; at the protein level this means replaces tyrosine at residue 54 with histidine — a missense variant. Submitter rationale: The Y54H variant in the CEACAM16 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The Y54H variant is not observed in large population cohorts (Lek et al., 2016). The Y54H variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret Y54H as a variant of uncertain significance.
