This variant lies in the TRIO gene (transcript NM_007118.4) at coding-DNA position 6314, where T is replaced by G; at the protein level this means replaces methionine at residue 2105 with arginine — a missense variant. Submitter rationale: The M2105R variant in the TRIO gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The M2105R variant is not observed in large population cohorts (Lek et al., 2016). The M2105R variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, in silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure/function. We interpret M2105R as a variant of uncertain significance.
