Uncertain significance — the classification assigned by GeneDx to NM_178012.5(TUBB2B):c.208C>T (p.Pro70Ser), citing GeneDx Variant Classification (06012015). This variant lies in the TUBB2B gene (transcript NM_178012.5) at coding-DNA position 208, where C is replaced by T; at the protein level this means replaces proline at residue 70 with serine — a missense variant. Submitter rationale: The P70S variant in the TUBB2B gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The P70S variant is not observed in large population cohorts (Lek et al., 2016). The P70S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. Additionally, the majority of missense variants in the TUBB2B gene are considered pathogenic (Stenson et al., 2014). We interpret P70S as a variant of uncertain significance.
