Likely pathogenic — the classification assigned by GeneDx to NM_001104631.2(PDE4D):c.1814A>G (p.Tyr605Cys), citing GeneDx Variant Classification (06012015): The Y605C variant in the PDE4D gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The Y605C variant is not observed in large population cohorts (Lek et al., 2016). The Y605C variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret Y605C as a likely pathogenic variant.
