This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 6384, deleting one base; at the protein level this means shifts the reading frame starting at leucine residue 2128, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This deletion of one nucleotide in ATM is denoted c.6384delG at the cDNA level and p.Leu2128PhefsX8 (L2128FfsX8) at the protein level. The normal sequence, with the base that is deleted in brackets, is CATT[delG]TACA. The deletion causes a frameshift which changes a Leucine to a Phenylalanine at codon 2128, and creates a premature stop codon at position 8 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay.
