This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 7791, deleting one base; at the protein level this means shifts the reading frame starting at glutamic acid residue 2598, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This deletion of one nucleotide in BRCA2 is denoted c.7791delA at the cDNA level and p.Glu2598LysfsX50 (E2598KfsX50) at the protein level. Using alternate nomenclature, this variant would be defined as BRCA2 8019delA. The normal sequence, with the base that is deleted in brackets, is GAAA[delA]GAAG. The deletion causes a frameshift which changes a Glutamic Acid to a Lysine at codon 2598, and creates a premature stop codon at position 50 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. We consider this variant to be pathogenic.
