NM_000179.3(MSH6):c.3226dup (p.Arg1076fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): This duplication of one nucleotide in MSH6 is denoted c.3226dupC at the cDNA level and p.Arg1076ProfsX17 (R1076PfsX17) at the protein level. The normal sequence, with the base that is duplicated in brackets, is GTGT[dupC]GCCC. The duplication causes a frameshift which changes an Arginine to a Proline at codon 1076, and creates a premature stop codon at position 17 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. We consider this variant to be pathogenic.
