Pathogenic — the classification assigned by GeneDx to NM_003072.5(SMARCA4):c.788dup (p.Gly264fs), citing GeneDx Variant Classification (06012015): This duplication of one nucleotide in SMARCA4 is denoted c.788dupC at the cDNA level and p.Gly264ArgfsX23 (G264RfsX23) at the protein level. The normal sequence, with the base that is duplicated in brackets, is TCCCC[dupC]AGGA. The duplication causes a frameshift which changes a Glycine to an Arginine at codon 264, and creates a premature stop codon at position 23 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. We consider this variant to be pathogenic.
