This variant lies in the PALB2 gene (transcript NM_024675.4) at coding-DNA position 1845, deleting one base; at the protein level this means shifts the reading frame starting at aspartic acid residue 616, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This deletion of one nucleotide in PALB2 is denoted c.1845delT at the cDNA level and p.Asp616MetfsX12 (D616MfsX12) at the protein level. The normal sequence, with the base that is deleted in brackets, is TACC[delT]GATG. The deletion causes a frameshift which changes an Aspartic Acid to a Methionine at codon 616, and creates a premature stop codon at position 12 of the new reading frame.
