Submitter rationale: This duplication of one nucleotide in BRCA1 is denoted c.5021dupT at the cDNA level and p.Thr1675HisfsX4 (T1675HfsX4) at the protein level. Using alternate nomenclature this variant would be defined as BRCA1 5140dupT. The normal sequence, with the base that is duplicated in brackets, is CACA[dupT]CACT. The duplication causes a frameshift which changes a Threonine to a Histidine at codon 1675, and creates a premature stop codon at position 4 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. We consider this variant to be pathogenic.
