Likely pathogenic — the classification assigned by GeneDx to NM_005687.5(FARSB):c.914G>A (p.Arg305Gln), citing GeneDx Variant Classification (06012015): The R305Q variant in the FARSB gene has been observed in internal GeneDx clinical exome sequencing data in association with lung disease and pneumothoraces, cirrhosis, intracranial aneurysms and calcifications, hypotonia, and connective tissue disease. The R305Q variant is not observed in large population cohorts (Lek et al., 2016). The R305Q variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. Therefore, we interpret R305Q as a likely pathogenic variant.
