Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003722.5(TP63):c.1063G>A (p.Asp355Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 355 of the TP63 protein (p.Asp355Asn). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of ectrodactyly‚Äìectodermal dysplasia‚Äìcleft palate (EEC) syndrome (PMID: 36071541; Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 544363). Advanced modeling performed at Invitae incorporating data from internal and/or published experimental studies (Invitae) indicates that this missense variant is not expected to disrupt TP63 function with a negative predictive value of 80%.
