Submitter rationale: The p.H662Y variant (also known as c.1984C>T), located in coding exon 9 of the BRCA1 gene, results from a C to T substitution at nucleotide position 1984. The histidine at codon 662 is replaced by tyrosine, an amino acid with similar properties. This alteration was observed in an individual diagnosed with breast cancer before the age of 40 (Loizidou M et al. Clin Genet. 2007; 71:165-70). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
