This variant lies in the QARS1 gene (transcript NM_005051.3) at coding-DNA position 1195, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 399 with lysine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 399 of the QARS protein (p.Glu399Lys). This variant is present in population databases (rs143072084, gnomAD 0.1%). This variant has not been reported in the literature in individuals affected with QARS-related conditions. ClinVar contains an entry for this variant (Variation ID: 544138).
