NM_007294.4(BRCA1):c.1945G>C (p.Glu649Gln) was classified as Uncertain significance by Quest Diagnostics Nichols Institute San Juan Capistrano, citing Quest Diagnostics criteria: The BRCA1 c.1945G>C (p.Glu649Gln) variant has been reported in the published literature in individuals with personal and/or family history of breast and/or ovarian cancer (PMID: 28525389 (2017), 16267036 (2005)), and described to be located in a region of the BRCA1 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). A functional study demonstrated that this variant had a neutral effect on protein function, however additional studies are needed to determine the global effect of this variant on protein function (PMID: 32546644 (2020)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant.
