Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_014391.3(ANKRD1):c.300G>C (p.Arg100Ser), citing Ambry Variant Classification Scheme 2023: The p.R100S variant (also known as c.300G>C), located in coding exon 3 of the ANKRD1 gene, results from a G to C substitution at nucleotide position 300. The arginine at codon 100 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
