Pathogenic for DICER1-related tumor predisposition — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_177438.3(DICER1):c.2370_2373dup (p.Leu792fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DICER1 gene (transcript NM_177438.3) at coding-DNA position 2370 through coding-DNA position 2373, duplicating 4 bases; at the protein level this means shifts the reading frame starting at leucine residue 792, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Loss-of-function variants in DICER1 are known to be pathogenic (PMID: 19556464, 21266384). This variant has not been reported in the literature in individuals with DICER1-related disease. For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Leu792Glufs*6) in the DICER1 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency).
