Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.328G>A (p.Val110Met), citing Ambry Variant Classification Scheme 2023: The p.V110M variant (also known as c.328G>A), located in coding exon 3 of the DICER1 gene, results from a G to A substitution at nucleotide position 328. The valine at codon 110 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
