NM_177438.3(DICER1):c.4103G>A (p.Arg1368His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1368H variant (also known as c.4103G>A), located in coding exon 21 of the DICER1 gene, results from a G to A substitution at nucleotide position 4103. The arginine at codon 1368 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
