NM_001365951.3(KIF1B):c.5225G>A (p.Arg1742His) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1696H variant (also known as c.5087G>A), located in coding exon 44 of the KIF1B gene, results from a G to A substitution at nucleotide position 5087. The arginine at codon 1696 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
