NM_001166108.2(PALLD):c.2303G>A (p.Arg768Lys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R264K variant (also known as c.791G>A), located in coding exon 4 of the PALLD gene, results from a G to A substitution at nucleotide position 791. The arginine at codon 264 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
