This variant lies in the FRRS1L gene (transcript NM_014334.4) at coding-DNA position 248, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 83 with serine — a missense variant. Submitter rationale: This sequence change replaces phenylalanine with serine at codon 134 of the FRRS1L protein (p.Phe134Ser). The phenylalanine residue is highly conserved and there is a large physicochemical difference between phenylalanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with FRRS1L-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
