Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_032737.4(LMNB2):c.700C>T (p.Arg234Trp), citing LabCorp Variant Classification Summary - May 2015: Variant summary: LMNB2 c.700C>T (p.Arg234Trp) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 0.00014 in 235148 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for disease-causing variants in LMNB2, allowing no conclusion about variant significance. c.700C>T has been observed in one individual affected with lipodystrophies (Varlet_2021). The report does not provide unequivocal conclusions about association of the variant with LMNB2-Related Disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 35011612). ClinVar contains an entry for this variant (Variation ID: 542428). Based on the evidence outlined above, the variant was classified as uncertain significance.
