Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8.6e-05 in 243180 control chromosomes. The observed variant frequency exceeds the estimated maximal expected allele frequency for disease-causing variants in SOS2. To our knowledge, no occurrence of c.1198G>A in individuals affected with SOS2-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
