Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001303256.3(MORC2):c.1812G>A (p.Glu604=), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MORC2 c.1812G>A (p.Glu604Glu) alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes a canonical 5' splicing donor site and one predicts the variant weakens the 5' donor site. However, these predictions have yet to be confirmed by functional studies. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. To our knowledge, no occurrence of c.1812G>A in individuals affected with MORC2-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
