Uncertain significance for Aicardi-Goutieres syndrome 7; Singleton-Merten syndrome 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022168.4(IFIH1):c.580G>C (p.Val194Leu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the IFIH1 gene (transcript NM_022168.4) at coding-DNA position 580, where G is replaced by C; at the protein level this means replaces valine at residue 194 with leucine — a missense variant. Submitter rationale: This sequence change replaces valine with leucine at codon 194 of the IFIH1 protein (p.Val194Leu). The valine residue is weakly conserved and there is a small physicochemical difference between valine and leucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with IFIH1-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
