Uncertain significance for Epidermolysis bullosa simplex 3, localized or generalized intermediate, with BP230 deficiency; Hereditary sensory and autonomic neuropathy type 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001723.7(DST):c.7940A>G (p.Tyr2647Cys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DST gene (transcript NM_001723.7) at coding-DNA position 7940, where A is replaced by G; at the protein level this means replaces tyrosine at residue 2647 with cysteine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with DST-related disease. This sequence change replaces tyrosine with cysteine at codon 2647 of the DST protein (p.Tyr2647Cys). The tyrosine residue is weakly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency).
