This variant lies in the ERCC4 gene (transcript NM_005236.3) at coding-DNA position 2591, where G is replaced by A; at the protein level this means replaces arginine at residue 864 with histidine — a missense variant. Submitter rationale: This sequence change replaces arginine with histidine at codon 864 of the ERCC4 protein (p.Arg864His). The arginine residue is moderately conserved and there is a small physicochemical difference between arginine and histidine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with ERCC4-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
