NM_006231.4(POLE):c.5108G>C (p.Cys1703Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C1703S variant (also known as c.5108G>C), located in coding exon 38 of the POLE gene, results from a G to C substitution at nucleotide position 5108. The cysteine at codon 1703 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
