Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.6033G>A (p.Met2011Ile), citing Ambry Variant Classification Scheme 2023: The p.M2011I variant (also known as c.6033G>A), located in coding exon 44 of the POLE gene, results from a G to A substitution at nucleotide position 6033. The methionine at codon 2011 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
