Submitter rationale: The c.4959dupT variant, located in coding exon 38 of the POLE gene, results from a duplication of T at nucleotide position 4959, causing a translational frameshift with a predicted alternate stop codon (p.H1654Sfs*40). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
