Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.4459A>C (p.Ile1487Leu), citing Ambry Variant Classification Scheme 2023: The p.I1487L variant (also known as c.4459A>C), located in coding exon 35 of the POLE gene, results from an A to C substitution at nucleotide position 4459. The isoleucine at codon 1487 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
