Uncertain significance for Charcot-Marie-Tooth disease dominant intermediate E; Focal segmental glomerulosclerosis 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022489.4(INF2):c.2984A>G (p.Asp995Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid with glycine at codon 995 of the INF2 protein (p.Asp995Gly). The aspartic acid residue is weakly conserved and there is a moderate physicochemical difference between aspartic acid and glycine. This variant is present in population databases (rs748234626, ExAC 0.002%). This variant has not been reported in the literature in individuals with INF2-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
