Pathogenic for Atrioventricular septal defect 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000008.11:g.(?_11703278)_(11758495_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the GATA4 gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Although a specific deletion of this gene has not been reported in the literature in individuals with GATA4-related disease, several larger deletions encompassing this gene have been reported in individuals and families with congenital heart disease (PMID: 22318994, 23239632, 23696316). Loss-of-function variants in GATA4 are known to be pathogenic (PMID: 12845333, 15235040). For these reasons, this variant has been classified as Pathogenic.
