This variant lies in the RAD51D gene (transcript NM_002878.4) at coding-DNA position 100, where G is replaced by A; at the protein level this means replaces alanine at residue 34 with threonine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with RAD51D-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces alanine with threonine at codon 34 of the RAD51D protein (p.Ala34Thr). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and threonine.
