This variant lies in the PIGN gene (transcript NM_176787.5) at coding-DNA position 47, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 16 with serine — a missense variant. Submitter rationale: This sequence change replaces phenylalanine with serine at codon 16 of the PIGN protein (p.Phe16Ser). The phenylalanine residue is moderately conserved and there is a large physicochemical difference between phenylalanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with PIGN-related conditions.
