NM_018979.4(WNK1):c.3623G>T (p.Ser1208Ile) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1460I variant (also known as c.4379G>T), located in coding exon 16 of the WNK1 gene, results from a G to T substitution at nucleotide position 4379. The serine at codon 1460 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
