Uncertain significance for Familial isolated pituitary adenoma — the classification assigned by St. Jude Molecular Pathology, St. Jude Children's Research Hospital to NM_004304.5(ALK):c.1517T>C (p.Leu506Pro), citing St. Jude Assertion Criteria 2020: The ALK c.1517T>C (p.Leu506Pro) missense change is absent in gnomAD v2.1.1, however this data may be unreliable due to poor data quality at this location (https://gnomad.broadinstitute.org/variant/2-29543646-A-G). Six of six in silico tools predict a benign effect of this variant on protein function (BP4), but to our knowledge these predictions have not been confirmed by functional assays. To our knowledge, this variant has not been reported in the literature in individuals with ALK-related neuroblastic tumor susceptibility.
