NM_002471.4(MYH6):c.3853G>A (p.Glu1285Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1285K variant (also known as c.3853G>A), located in coding exon 25 of the MYH6 gene, results from a G to A substitution at nucleotide position 3853. The glutamic acid at codon 1285 is replaced by lysine, an amino acid with similar properties. This variant was reported in individual(s) with features consistent with hypertrophic cardiomyopathy (HCM) (Janin A et al. Mol Diagn Ther, 2021 May;25:373-385). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
