This variant has been observed in individual(s) with oculodentodigital dysplasia (PMID: 19338053, Invitae). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (ExAC no frequency). This sequence change replaces lysine with asparagine at codon 102 of the GJA1 protein (p.Lys102Asn). The lysine residue is highly conserved and there is a moderate physicochemical difference between lysine and asparagine. For these reasons, this variant has been classified as Pathogenic.
