Uncertain significance for Ehlers-Danlos syndrome, dermatosparaxis type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014244.5(ADAMTS2):c.1208C>T (p.Ala403Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with valine at codon 403 of the ADAMTS2 protein (p.Ala403Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals affected with ADAMTS2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
