Uncertain significance for Brugada syndrome 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_201596.3(CACNB2):c.710A>C (p.Asn237Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with threonine at codon 183 of the CACNB2 protein (p.Asn183Thr). The asparagine residue is moderately conserved and there is a small physicochemical difference between asparagine and threonine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CACNB2-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
