Uncertain significance for Aortic aneurysm, familial thoracic 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001613.4(ACTA2):c.196A>C (p.Ile66Leu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ACTA2 gene (transcript NM_001613.4) at coding-DNA position 196, where A is replaced by C; at the protein level this means replaces isoleucine at residue 66 with leucine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with leucine at codon 66 of the ACTA2 protein (p.Ile66Leu). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and leucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with ACTA2-related disease. This variant is not present in population databases (ExAC no frequency).
