Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004064.5(CDKN1B):c.211G>C (p.Glu71Gln), citing Ambry Variant Classification Scheme 2023: The p.E71Q variant (also known as c.211G>C), located in coding exon 1 of the CDKN1B gene, results from a G to C substitution at nucleotide position 211. The glutamic acid at codon 71 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
