Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_017636.4(TRPM4):c.3130A>T (p.Ser1044Cys), citing Ambry Variant Classification Scheme 2023: The p.S1044C variant (also known as c.3130A>T), located in coding exon 20 of the TRPM4 gene, results from an A to T substitution at nucleotide position 3130. The serine at codon 1044 is replaced by cysteine, an amino acid with dissimilar properties. This alteration has been reported in a subject with complete heart block and the same authors noted a possible impact on protein function (Bianchi B et al. Front Physiol, 2018 Mar;9:177). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
