Pathogenic for Autosomal recessive limb-girdle muscular dystrophy — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_213599.3(ANO5):c.1120-1G>A, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the ANO5 gene (transcript NM_213599.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1120, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: ANO5 c.1120-1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of ANO5 function. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8e-06 in 250156 control chromosomes. c.1120-1G>A has been reported in the literature as homozygous in individuals affected with Autosomal Recessive Limb-Girdle Muscular Dystrophy and Miyoshi myopathy (e.g. Papadopoulos_2017). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
