Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.12:g.(?_161785752)_(161973437_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 6-7 of the PRKN gene. This deletion is out-of-frame, and is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Loss-of-function variants in PRKN are known to be pathogenic (PMID: 10072423, 20301651, 22956510). A similar copy number variant has been observed in individuals with early-onset Parkinson's disease (PMID: 10939576, 12764051, 24677602, 25833766). For these reasons, this variant has been classified as Pathogenic.
