NM_024422.6(DSC2):c.1996G>A (p.Val666Ile) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V666I variant (also known as c.1996G>A), located in coding exon 13 of the DSC2 gene, results from a G to A substitution at nucleotide position 1996. The valine at codon 666 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, isoleucine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
