Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000548.5(TSC2):c.775-5C>G, citing Sema4 Curation Guidelines: The TSC2 c.775-5C>G variant has not been reported in literature to our knowledge. This variant was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654), but has been reported in ClinVar (Variation ID: 536051). Algorithms developed to predict the effect of sequence changes on RNA splicing do not suggest a negative effect, though these predictions have not been confirmed by functional studies. There is no indication that this variant causes disease, but the evidence is insufficient currently to prove that conclusively. Thus, the clinical significance of this variant is currently uncertain.
