NM_000548.5(TSC2):c.4300G>T (p.Gly1434Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1434C variant (also known as c.4300G>T), located in coding exon 33 of the TSC2 gene, results from a G to T substitution at nucleotide position 4300. The glycine at codon 1434 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
